  • Can Flow-Through and Bound Peak Areas be Utilized to Calculate Depletion Efficiency?
No – the change in flow rates as well as variability of proteins response to UV makes using the peak area/absorbance not accurate. Generally, one can overlay the chromatograms to inspect reproducibility run to run to ensure flow-through peaks and bound peaks are not changing. But a direct relationship to peak area or peak height is not accurate for calculating efficiency of removal.
