It is our goal to remove the targeted proteins and minimal amount of non-target ones. Polyclonal antibodies are used to ensure binding specificity. We have evaluated column binding specificity and found that the Multiple Affinity Removal System is the most targeted high-abundant protein removal device compared to conventional protein removal products.
- Tryptic in-gel digestion of bound proteins from the eluted fraction and LC/MS/MS analysis indicates that Apolipoprotein-A1, Complement C3, and Complement C4 are the only non-target proteins detected in the bound fraction. Binding of these non-targeted proteins represents only a small fraction of the flow-through quantities.
- ELISA results indicate that > 99% of target proteins have been removed by the affinity column.
- Mobile phase composition (Buffer A) for binding and washing has been formulated to minimize removal of proteins not targeted by the antibodies.
- In any case, if low-abundant proteins are associated with the captured high-abundant proteins, they can be easily recovered by collecting them in the bound fraction and can then be analyzed further.
