Rapid Analysis of Vitamin D in Serum Using Triple Quadrupole LC/MS
Abstract:
A rapid method for the quantification of 25-OH D2 and 25-OH D3 in serum was developed
using the Agilent 1200 Series Rapid Resolution LC (RRLC) system coupled with an Agilent
6430 Triple Quadrupole LC/MS with multimode ionization (MMI). Solid phase extraction
minimizes matrix interferences and ion suppression, while enabling automation.
Excellent linearity of quantification (R2 > 0.99) was obtained in the range of 5 to 100 ng/mL, with
run times of only 5 min, and day-to-day precision was well below 10%. The limits of detection
(LOD) and limits of quantification (LOQ) were 2 ng/mL and 3 ng/mL respectively, in serum.
