Reproducibility of the HPLC-Chip for 17 peaks in a BSA tryptic digest sample (N=69).
A 50 fmol of BSA digest was injected 69 times on the HPLC-Chip in a sequence using a fast chromatographic method with MS acquisition only and 17 peaks were evaluated for retention time reproducibility. The above table shows the results for the selected peaks, with an average %RSD of 0.27 (average SD of 0.01) across all runs for the 17 peaks. In a second experiment, a mixture of four peptides (angiotensin I, LHRF, neurotensin, and substance P) was injected 200 to 300 times on three different chips. No deviation for retention time could be observed run-to-run or even within 10 runs. The mean average deviation over hundreds of runs was always less than 0.2 min. The same sample was analyzed to determine chip-to-chip variation. For a series of 35 Protein ID chips (G4240-62001), standard deviation for retention times was 0.10 min for LHRF and 0.26 min for substance P.
