In nanoflow chromatography, a major source of peak dispersion originates from the connecting capillaries and fittings. This becomes critical when the ratio of peak volume to connecting capillary dead volume becomes too small. For a 75 µm column on a conventional nanospray system, the relative contribution of all connecting capillary volumes can reach up to 45 % of the peak volume causing significant dispersion and band broadening. On the HPLC-Chip, the nanocolumn and nanospray emitter are connected by a short channel with minimal dead volume that eliminates peak dispersion and ensures uncompromised chromatographic performance.
